We will focus on orthologues of human genes, the products of which have been associated with serious diseases. Since for ethical reasons, functional studies in humans are virtually impossible and many analyses in related mammals are cost prohibitive and/or limited by a number of factors, the role of model organisms in the study of complex human diseases is quickly growing. In frame of this project we want to dissect the functions of selected proteins in model organisms Trypanosoma brucei, Caenorhabditis elegans and Drosophila melanogaster. Using cutting edge methods of genetic manipulation (RNA interference, knock-in, complementation etc.) we will particularly concentrate on functional analysis of i/ proteins involved in iron-sulfur cluster synthesis (e.g. frataxin), ii/ nuclear receptors (e.g. NHR-25), iii/ transcriptional cofactors (e.g. MBF1) and iv/ proto-oncoproteins"@en . .
