Producing adequate amounts of recombinant proteins is a key problem in using this technology. We aim to further the development of a novel, high-yield, inducible protein expression system capable of producing post-translationally modified eukaryotic proteins. The biology of kinetoplastids will be exploited by using non-pathogenic flagellate Leishmania tarentolae as a protein expression platform. A proteomic approach will be used to identify regulatory elements in the L. tarentolae genome that stabilize mRNA and enhance translation initiation in response to signals such as chemical, cold or heat shocks. These elements are vital for the development of a tightly inducible protein expression system. The suitability of the developed system for structural and functional genomics will be addressed by overexpressing human proteins as well as such para-sites L. major and T. brucei."@en .
