"The main aim of the project is application of modern, high selective and extremely sensitive analytical tools for study of the hormone function in plant growth and development. We will use the phytohormone profiling approaches (targeted metabolomics) for screening of all phytohormone groups in minute amount of fresh plant material. Development of new isolation methods (extraction and purification) from one sample will be combining with optimization of analytical methods \u2013 ultra high performance chromatography and tandem mass spectrometry. Preparation of stable isotope-labelled internal standards is also part of the project to simultaneously profile the majority of known plant growth regulators (auxins, cytokinins, gibberellins, abscisic acid, brassinosteroids and jasmonates). We will validate the method by profiling the phytohormone metabolome in different tissues (roots, shoots and flowers) from various Arabidopsis thaliana ecotypes and over-producing mutant lines.
