"The project is aimed in development of an expression system for preparation of lytically active enzymes exploitable as antimicrobial preparations for therapy and control of staphylococcal infections. Genes of staphylococcal bacteriophages with broad host range encoding lytic enzymes will be optimized by gene engineering methods to enhance their lytic activity. A set of gene construct as well as suitable expression system for protein production will be established. To enhance lytic effect of the enzymes, the fused proteins will be designed in which the phage species-specific peptidoglycan-binding domain SH3b will be combined with catalytic domains derived from different phage lytic enzymes. The resulting recombinant lysins will be characterized biochemically and atomic structure of selected protein domains responsible for interaction with pathogen cells will be defined.
