"Within this project we will continue our previous research focused on developing electrochemical tools for DNA hybridisation detection. We will jodidy oligonucleotides with various tags (primarily metal complexes), study thein electrochemical behaviour and use them as signalling probes in DNA hybridization. OsO4 complexes will be used as chemical probes for unpaired and mispaired bases in DNA heteroduplexes. This principle will be utilized in a new technique of detection of point mutations in DNA. For our purposes we will optimise conditions for manipulating with DNA duplexes containing mismatches and AP sites and conditions for their electrochemical and/or immunodetection. We will employ double-surface strategy of DNA hybridisation detection, in which the hybridisation occurs on separated surfa\u0159e - paramagnetic beads.
