"The aim of proposed project is the development of highly sensitive and selective analytical methods with the intention of use for diagnostic determination of biological markers ascorbic and dehydroascorbic acid and also neopterin and its derivatives in biological matrices. Therefore hyphenation of Ultra Performance Liquid Chromatography (UPLC) and mass spectrometry using triple quadrupole analyzer was chosen as optimal method. This technique is convenient by its high sensitivity and selectivity and thus it is used for quantitation of substances present in trace amounts even at complicated matrices as biological materials. Methods of chromatographic UPLC separation of tested compounds will be optimized together with the conditions of mass spectrometric detection. Isolation procedure of target analytes from biological materials will be optimized thereafter with the aim to get the highest possible recovery and method precision.
