"The aim of this project is to provide detailed characterization of the newly discovered chicken cellular receptor (Tvc), a member of the butyrophilin superfamily, which is employed by avian sarcoma-leukosis viruses of subgroup C (ASLV-C) to gain entry into cells. We will attempt to identify the binding site for retroviral envelope glycoprotein in the Tvc protein and the binding capacity of chicken and duck Tvcs for different retroviruses of subgroup C. To get insight into the physiological function ofthe Tvc receptor we will pursue isolation of both, a natural ligand and a protein associated with the Tvc intracellular B30.2 domain. An integral part of this study will be determination of viral genome regions involved in the ASLV-C pathogenicity manifested in vivo by wasting disease as well as deciphering the role of the Tvc receptor in retroviral cythopathogenicity displayed in vitro.
