"The proposed project is aimed at obtaining information about variations in the pattern of the studied genes expression during the inductive pretreatment for androgenesis. The effects of temperature treatment, and osmotic, and nutritional stress on the pollen embryogenesis induction will be analysed in relation to the genotype of donor plant, and to the stages of microspore and pollen development. RT-PCR, northern-blot hybridisation analyses, PAGE, 2-D, and micro IEF protein electrophoresis will be used to characterise both qualitative and quantitative differences. The gene specific primer sets will be designed from the known , of genes of interest (LEA, HSP, HSP16.9, RKIN1, BKIN12, PKABA1 and Gli) and/or published PCR primers and will be optimalized for RT-PCR. Project will enable to obtain new pieces of information concerning the activity of the studied genes during the process of pollen embryogenesis in cereals in relation to the applied stress treatment.
