"Using the model tick Ixodes ricinus, we have recently demonstrated that tick intestinal digestion of host blood is based on gut-associated cysteine and aspartic peptidases. The peptidase spectrum is similar to the digestive cascade/network of blood feeding platyhelminths and nematodes. The aspartic protease cathepsin D (IrCD) presents one of the apical enzymes in the hemoglobin degradation cascade in the tick gut. The aims of the proposed project are: active recombinant IrCD, biochemical characterization, novel data on IrCD substrate specifity, high resolution IrCD 3-D structure and IrCD-RNAi knock-down phenotype. The overall goal is to confirm the role of IrCD in the hemoglobinolytic cascade and to test the putative potential to produce hemoglobin derived anti-microbial peptides.
