"Nuclear receptors play vital roles in development and reproduction of all metazoans. The genome of C. elegans encodes an extreme number of 270 nuclear receptors (NHR), of which less than 10 have known functions. One is NHR-25, whose orthologs mediate molting and metamorphosis in Drosophila, and gonadal differentiation and steroidogenesis in mouse. We have shown the requrement of NHR-25 for C. elegans embryogenesis, molting, and gonad and vulva differentiation. However, the mode of NHR-25 action and its target fenes remain unidentified. Here we utilize our nhr-25 mutant and RNA interference to dissect the role of NHR-25 in cuticle replacement and gonad differentiation. Using transgenic reporters we will identify targets regulated by NHR-25 among candidate genes involved in gonadal signaling and, by analogy with mammals, in steroidogenesis.
