Excessive adenosine is harmful, as in the case of adenosine deaminase (ADA) deficiency causing severe combined immunodeficiency in humans. Since the mechanism of such effects is often unclear, we have established a Drosophila model for ADA deficiency which will allow new genetic tools to be applied to the problem. We propose to establish a transcription reporter for the gene encoding ADGF-A, which is the main regulator of adenosine levels in Drosophila, and use it to study the role of adenosine in immunomodulation and its relationship with the NF-kB signaling cascade. We will investigate the spatiotemporal regulation of adenosine levels and the potential role of adenosine as a developmental signal.
