"The primary enamel knot (PEK) represents a signalling centre of developing teeth responsible for transition of molar tooth germs from bud to cap and bell stage. Therefore, PEK is considered as an important cell population with high impact on final tooth crown morphology and tooth size. PEK cells are eliminated by apoptosis, BMP/FGF interplay is suggested as the initiating step. However, not much is known about the following pathways. Cell death in PEK was shown as caspase dependent, nevertheless, alternative death (necrosis, autophagy) after caspase inhibition might be involved. This project aims to connect research done in frame of GAAV2005-2007 and recent challenges concerning PEK cell death. The PEK cells will be studied in context of the whole tooth organ using modern explant culture approach. Moreover, novel specific techniques for analysis of limited cell populations will be designed and exploited. Modulations of PEK cell death may be further applied in molecular dentistry."@en . .
