"In this project we propose to employ chemical probing of DNA structure, in connection with biochemical methods and electrochemical detection, in analysis of DNA damage. Monitoring of DNA-damaging pollutants in the environment, together with analysis of damage to cellular DNA, belong to important steps in prevention of serious diseases such as cancer. We propose to use osmium tetroxide complexes (Os,L) in detection of DNA damage induced by different chemical and physical agents. DNA-Os,L adducts will be detected electrochemically, or using enzyme-linked immunoassay with electrochemical detection. In a new approach, magnetic beads will be used as carrier surface for DNA exposed to DNA damaging agents and/or to capture and purify damaged DNA from reaction mixtures (cell lysates) for subsequent electrochemical analysis. To gain the best response, currently used elekctrochemical techniques will be improved and optimized, and simple electrochemical DNA biosensors will be designed."@en . .
