"The aim of the project is to develop a new diagnostic system for ultrasensitive detection of selected rare proteins in body fluids with dynamic range up to 8 orders of magnitude. The test is based on binding of the protein under study to a monoclonal antibody (mAb) immoblized together with thiolated DNA oligonucleotides on gold nanoparticles (NP). Next, the complex of the protein bound to NP is anchored to magnetic immunoparticles. Thiolated DNA oligonucleotides are then released and serve as initiating and limiting DNA primers in quantitative real-time PCR. Utility of the method will be verified by detection of rare proteins in body fluids in patients with neuropsychiatric diseases and for detection of rare cytokines. The project will include testing of newly prepared mAb for their performance in nano-PCR. We assume that nano-PCR will be 6-8 orders of magnitude more sensitive than ELISA and will allow development of diagnostic methods of new generations."@en . . . "42799"^^ . .
