"In this project we intend to prove that tick alpha-2-macroglobulin (IrAM) mediates the pathogen transmission by the hard tick Ixodes ricinus. We will study the interaction of IrAM with two tick pathogens: a model Gram-negative bacteria Chryseobacterium indologenes and the Lyme disease spirochetes Borrelia burgdorferi. The antibodies raised against a recombinant fragment of IrAM will be used to visualize binding of IrAM to both pathogens by immunofluoerscent microscopy. Suitable in vitro and in vivo assays will be designed to ascertain how IrAM inactivation (by methylamin) or silencing of its gene (by dsRNA inteference) will influence the ability of tick hemocytes to phagocytose bacteria and spirochetes. Since IrAM is a broad spectrum protease inhibitor, we will investigate its interaction with proteases associated with pathogen surface.
