"This project has four related but sufficiently independent parts: Part I uses bisulfite sequencing and chromatin IP on human samples (biopsies, and stable cell lines) to address natural epigenetic modifications of the six potentially most active human L1 elements. It will also study transcription and its regulation at these six loci. Part II employs bisulfite sequencing of mouse DNA (different tissues and developmental stages) to map epigenetic reprogramming of the transgenic human L1 element and its retrotransposed de novo insertions during the mouse life cycle. Part III focuses on using L1 retrotransposition model in cell culture to study mechanisms involved in the control of L1 silencing and to analyze possible inhibitory effects of simultaneous expression of both RNA strands. Part IV will address (in cell culture) whether several L1 inverted repeats dispersed in the human genome generate double-stranded RNA, which in turn may contribute to L1 silencing."@en .
