JH regulates insect development by preventing metamorphosis in larvae that have not yet reached the appropriate stage. While four decades have passed since the sesquiterpenoid JH structure was discovered, we still do not know how this hormone works, what receptor and signaling pathway it uses. Such knowledge is important because insects are the most numerous of animals on the Earth, and metamorphosis contributed to their success. Perturbing of JH signaling is a good way to control insect pests. The exquisite model beetle Tribolium has enabled us to show that JH regulates metamorphosis through a Methoprene-tolerant (Met) protein of the bHLH-PAS family. Here we propose to determine how the transcription factor Met transduces the JH signal and whether it serves as a JH receptor. We plan to disclose the molecular structure and interactions of Met, its binding and activation by JH, and to identify its target genes."@en . . .
