Much research has focused on the core RNA editing machinery, while other accessory factors, proteins implicated in this process, are in various stages of understanding. We have recently shown, also by solving its crystal structure, that the MRP protein complex has a multifarious role in RNA metabolism. Preliminary data suggest a role in RNA editing and perhaps in other RNA processing for TbRGG1 and REAP1. We propose to further the understanding of these genes via the functional genetic tools available in Trypanosoma brucei to i/ silence specific genes to determine an effect on RNA editing or other RNA processing events; ii/ knock-in tagged wild-type or mutant versions of these genes to find protein partners and determine features that facilitate this binding; iii/ over-express the above and newly discovered proteins for in vitro RNA binding assays and structural studies."@en .
