"CD161, C-type lectin-like receptor is expressed on immune cells with cytotoxic effector function (NK, NKT, monocytes). Our previous findings demonstrated that saccharides with N-acetyl-D-glucosamine (GlcNAc) moiety represent the candidate ligands. The aim of current project focuses on the identification of cell population/function affected by synthetic GlcNAc-terminated glycoconjugates (GC) with linear or dendritic structure. We plan to assess the dynamics of CD161 protein expression upon interaction with GCs on cellular, subcellular and gene transcription. FACS analysis and laser scanning cytometry using fluorochrome labeled GCs will be combined with functional assays. The project involves analysis of signal transduction pathways mediated by G-proteins, and their possible crosstalk with Src family kinases. In vivo application of GCs with characterization of cytotoxic lymphocytes could answer the question of CD161 participation in their maturation, migration, and effector function."@en . "0"^^ . . . .
