V r\u00E1mci tohoto projektu bude na\u0161e pr\u00E1ce zam\u011B\u0159ena na n\u00E1sleduj\u00EDc\u00ED hlavn\u00ED t\u00E9mata: (1) Dal\u0161\u00ED zdokonalen\u00ED detekce zlom\u016F v DNA s vyu\u017Eit\u00EDm p\u0159edev\u0161\u00EDm stu\u0165ov\u00FDch filmov\u00FDch elektrod (MFE), a optimalizace anal\u00FDzy DNA na MFE. (2) Modifikace d\u0159\u00EDve vyvinut\u00E9 metody v techniku umo\u017E\u0148uj\u00EDc\u00ED detekci zlom\u016F v eukaryotick\u00E9 chromozom\u00E1ln\u00ED DNA. (3) Detekce po\u0161kozen\u00ED baz\u00ED v DNA s vyu\u017Eit\u00EDm enzym\u016F z\u00FA\u010Dastn\u011Bn\u00FDch v opravn\u00FDch procesech DNA. Enzymy budou vyu\u017Eity jak pro \u0161t\u011Bpen\u00ED po\u0161kozen\u00E9 DNA v roztoku, tak na povrchu elektrody. "It is proposed to develop a fast response, simple, selective and versatile electrochemical biodetector (biosensor) for DNA damage (both in vitro and in cells) and DNA damaging agents. A procedure developed in our laboratory which allows to detect DNA strand breaks using mercury electrodes modified with supercoiled DNA will represent the ground of this research. Within this projects, our efferts will be focused mainly the following topics: (1) Improvement of detection of DNA strand breaks using mainlymercury film electrodes (MFE, and optimization of DNA analysis with the MFE. (2) Modification of the previously developed protocol into a technique allowing sensitive detection of DNA cleavage using eukaryotic chromosomal DNAs. (3) Detection of damage toDNA bases using DNA repair enzymes. The enzymes will be applied both in solution and at the electrode surface.
