"The aim of this project is the development of novel techniques of DNA labeling and their utilization in DNA electrochemical analysis and biosensor design. The DNA labeling will involve (1) synthesis of nucleoside triphosphate conjugates (modified at nucleobase) with redox-active moieties using aqueous cross-coupling reactions, and (2) their incorporation to DNA by enzymes. Methodology of construction of DNA probes bearing redox markers at specific positions will be developed. Electrochemical features of the modified nucleotides and nucleic acids, as well as the influence of DNA structure on electrochemical responses of the conjugate markers, will be studied. Based on these investigations, we will choose labels and analytical procedures suitable for efficient analysis of nucleotide sequences, with emphasis on applications in molecular diagnostics of severe diseases.
