"We propose to study synthetic oligonucleotides (ODN) using electrochemical techniques at mercury and carbon electrodes. Information about the influence of sequence and length of the ODN on their electrochemical behavior will be obtained. Behavior of ODN adopting various secondary structures (besides duplex form, hairpins, triplexes or quadruplexes) will be studied. Transitions between them are accompanied by changes of accessibility of certain base residues influencing the electrochemical signals. Formation of the alternative structures occurs under different conditions (pH, presence of metal ions); most of the experiments will be therefore performed in medium exchange mode enabling to change conditions under which the ODN are adsorbed at electrodes while keeping conditions of the measurements constant. Besides improvement of basic knowledge in the field of DNA electrochemistry, one of the project goals is to propose novel analytical approaches applicable in practical DNA diagnostics."@en . "2"^^ .
