We show that individual cognate pMHC complexes displayed by dendritic cells (DCs), as compared with nonprofessional APCs, are far better in productively triggering Ag-specific TCRs independently of conventional costimulation. As we further show, this is accomplished by the unique ability of DCs to robustly activate the Src family kinases (SFKs) Lck and Fyn in T cells even in the absence of cognate peptide. Instead, this form of SFK activation depends on interactions of DC-displayed MHC with TCRs of appropriate restriction, suggesting a central role of self-pMHC recognition. DC-mediated SFK activation leads to %22TCR licensing,%22 a process that dramatically increases sensitivity and magnitude of the TCR response to cognate pMHC. Thus, TCR licensing, besides costimulation, is a main mechanism of DCs to present Ag effectively. Komplexy pMHC na povrchu dendritických buněk (DC) jsou ve srovnání s jinými typy neprofesionálních APC mnohem účinnější ve stimulaci antigenně-specifických T lymfocytů, nezávisle na konvenční kostimulaci. To je způsobeno unikátní schopností DC robustně aktivovat v T lymfocytech kinasy skupiny Src (SFK) Lck a Fyn, a to dokonce v nepřítomnosti specifického pMHC. Tato forma aktivace SFK závisí na interakcích MHC na povrchu DC s TCR, což poukazuje na ústřední úlohu rozpoznání self-MHC. Aktivace SFK mediovaná DC vede k „TCR licensing“, což je proces, který dramaticky zvyšuje citlivost a velikost odpovědi TCR na pMHC. TCR licensing je tedy kromě kostimulace hlavním mechanismem zodpovědným za efektivní prezentaci antigenu pomocí DC.
