Aurora B kináza je tzv. chromosomální „passenger“ protein, který je nezbytný pro řadu procesů probíhajíchích během mitózy. Jeho aktivita je regulována spojením s dalšími „passenger“ proteiny, INCENP a Survivin, a jeho fosforylací na Thr-232. V této práci, byla studována exprese a fosforylace (Thr-232) Aurory B kinázy během meiotického zrání prasečích oocytů spolu s fosforylací histonu H3 a v korelaci s kondenzací chromosomů. Ukázalo se, že fosforylace histonu H3 na Ser-10, ale ne na Ser-28, koreluje s postupnou kondenzací chromosomů během zrání oocytů; fosforylace na Ser 10 začíná v okolí doby rozpadu jaderné membrány, s maximální aktivitou v metafázi I, zatímco fosforylace na Ser-28 se během zrání oocytů výrazně nemění. Po ošetření oocytů 50 mM Butyrolactonem I (BL-I), inhibitor cyklin dependentních kináz, či cykloheximidem ( 10mg/ml) – inhibitor proteosyntézy, dochází k zastavení oocytu ve fázi tzv. zárodečného váčku (germinal vesicle – GV), ve které zůstává neporušena jaderná membrána, nicm... In this study, we have examined expression and phosphorylation (Thr-232) of Aurora B kinase during meiotic maturation of pig oocytes in correlation with histone H3 phosphorylation and chromosome condensation. We show that histone H3 phosphorylation on Ser-10, but not on Ser-28, correlates with progressive chromosome condensation during oocyte maturation; Ser-10 phosphorylation starts around the time of the breakdown of the nuclear envelope, with the maximal activity in metaphase I, while Ser-28 phosphorylation does not significantly change in maturing oocytes. Treatment of oocytes with 50 mM Butyrolactone I (BL-I), an inhibitor of cyclin-dependent kinases, or cycloheximide (10mg/ml), inhibitor of proteosynthesis, results in a block of oocytes in the germin...
