S. cerevisiae mutant strains lacking the genes encoding alkali-metal-cation exporters (NHA1, ENA1, ENA2, ENA5, TOK1) and potassium importers (TRK1, TRK2) have been constructed. The TRK1 gene has been substituted by the LEU2 gene; the three ENA genes, TOK1 and TRK2 genes have been replaced by the kanMX cassette. Finally, the LEU2 and kanMX cassettes from all strains were replaced by cre-loxP system leaving a single loxP site.
