The ABCB1 gene encodes P-glycoprotein, a member of the multi-drug resistance protein subfamily B, which plays a role in the cell efflux of imatinib mesylate (IMA). Although some conflicting results have been reported, it is generally accepted that overexpression of the ABCB1 gene leads to the upregulation of P-glycoprotein (i.e.ABCB1) and the subsequent increase of IMA efflux, which has been postulated to be an important mechanism of drug resistance in CML (1). Despite an abundance of literature that discusses ABCB1 biology, only limited data exist that have explored the relationship between ABCB1 expression or activity and the prediction of treatment response in patients with chronic myelogenous leukemia (CML) (2-5).
