Claw diseases in dairy cows are the third most important disease group, with laminitis being the primary disorder. To date not all of the causes of laminitis are fully understood. However, it is known that the postpartum period is the most critical time. With the onset of lactation, nutrient requirements significantly increase and this results in the need for major changes to the diet. These changes may lead to the development of ruminal acidosis and consequently increased production of histamine in the rumen. Histamine exerts a local effect on microcirculation in the claw. In vivo and in vitro studies have previously demonstrated that claw keratinocytes are able to produce both immunoreactive enzyme matrix metalloproteinase-2 (MMP-2), the expression of which is increased during laminitis, and cytokine interleukine-1 (IL-1), which in turn triggers the inflammatory cascade. The aim of this study was to determine the influence of different levels of histamine acting for different lengths of time on bovine keratinocytes, and subsequent determination of MMP-2 and IL-1 in the medium.
