RAPD and SSR markers were used for genetic diversity evaluations among 15 genotypes selected from the genus Prunus L. Altogether 40 RAPD primers and 21 primer pairs designated for microsatellite loci were applied on the whole group of genotypes. The RAPD primers and SSR loci with the highest degree of observed polymorphism were selected. Three approaches were used to generate DNA-based genetic markers--RAPD, SSR with separations on PAGE, SSR with separations on MetaPhor agarose. An analysis of each approach was statistically summarized and the obtained parameters were compared. On the basis of the data obtained by individual methods, the relevant dendrograms of genetic similarities among genotypes were constructed. The Mantel tests between individual similarity matrices showed a statistically significant correlation between the RAPD- and SSR-based similarities if computed for the whole group of analyzed genotypes. Genetické markery byly připraveny pomocí tří přístupů - RAPD, SSR při separaci na agaróze Metaphor a SSR při separaci na polakrylamidovém gelu. Byly porovnány výsledky získané na základě aplikace těchto tří přístupů. Vzhled dendrogramu genetické podobnosti se teoretickým předpokaldů nejvíce blížil v případě RAPD metody.
