Ellipticine (5,11-demethylovaný-6H-pyridol (4,3-b) karbazolu) je rostlinný alcaloid izolován z Apocyanaceae rodiny (např. Ochrosia borbonica, Excavativa cocinea). Tato sloučenina a některé jeho deriváty vykazují výrazný protinádorový účinnek. Protinádorový mechanismus ellipticinu spočívá v DNA interiakcí a / nebo inhibice topoizomerázy II. Bylo prokázáno, že ellipticine aktivován pomocí cytochromu P450 je schopen kovalentně vázat na deoxyguanosine ve struktuře DNA. Cílem této práce bylo studium interakcí ellipticine a DNA pomocí miniaturizovaný elektrody. V našich experimentech je vyrobena miniaturizovaný senzor. Prvním krokem bylo studium ellipticine je (0,5, 1 uM) elektrochemické chování pomocí cyklické voltametrie a obdélníkovou vlnou. Z voltammogramů získané oxidační signály v potenciál asi 600 mV byly pozorovatelné. Dále elektrochemické vlastnosti dsDNA byly studovány. Mez detekce dsDNA byla nižší než 1 ug / ml a jednotné základy byly odlišeny. V následujících pokusech interakce ellipticine a Ellipticine (5,11-demethyl-6H-pyridol(4,3-b)carbazole) is a plant alcaloid isolated from Apocyanaceae family (e.g. Ochrosia borbonica, Excavativa cocinea). This compound and some its derivatives exhibit significant antitumour activity. Mechanism of ellipticine antitumour consists in DNA intercalation and/or topoisomerase II inhibition. It was shown that ellipticine activated by cytochrome P450 is able to bind covalently to deoxyguanosine in DNA structure. The aim of this work was to study of interaction of ellipticine and DNA using miniaturized electrodes. In our experiments the fabricated carbon paste printed electrodes modified with carbon nanotubes were used. The first step was to study of ellipticine's (0.5, 1 uM) electrochemical behavior by using cyclic and square wave voltammetry. In voltammograms obtained the oxidation signals in potential of about 600 mV were observable. Further the electrochemical properties of dsDNA were studied.
