To carry out solid-phase peptide synthesis (SPPS), the first building block, an amino acid, needs to be temporarily attached to the insoluble support. The immobilization occurs via a linker- a chemical moiety that allows temporary linking of the first amino acid to solid support and release of target peptide at the end of synthesis. In this chapter, we reviewed diversity of linkers developed for SPPS.
