The method for estimation of 37 naturally occurring cytokinins is based on an immunoaffinity purification (IAC) and a baseline chromatographic separation of 20 cytokinins (trans/cis-zeatin, dihydrozeatin, isopentenyladenine, benzyladenine, meta/ortho-topoline groups) on reversed-phase column and analysis by high-performance liquid chromatography-mass spectrometry (HPLC/MS). When low cone voltage (20 V) was applied, all studied cytokinins were determined in aqueous methanol as dominant quasi-molecular ions of [M+H]+ with limits of detection between 10 and 50 fmol and with linearity ranges of calibration between 25 fmol and 100 pmol. The method was validated by deuterium or 15N labeled standards (isoprenoid and aromatic), added during extraction. Two different extraction procedures (70% ethanol or Bieleski buffer) have been tested.
