The separation, identification and quantitative analysis of phenolic acids are necessary because of their importance as non-steroid antiphlogistic drugs, semi-products of biosynthesis of aromatic amino-acids in plants and natural products with antioxidative and anticarcinogenic abilities. A method was developed for LC/MS analysis of eighteen phenolic acids in plant material after methanolic extraction. Different stacionary (C8, C18, Phenyl-Hexyl) and many mobile phases were tested during an optimisation process. Two fundamentally different atmospheric pressure ionization interfaces (chemical and electrospray ionizations) were also compared in terms of ionization effectivity and limits of detection. The most suitable method for analysis of phenolic acids was carried out after combining Fenyl-Hexyl column, gradient elution (acetonitrile - 5 mmol.L-1 HCOOH) and negative mode electrospray ionization.
