Rapid and sensitive method for analysis of 12 Cytokinins in mg amount of plant tissue has been developed. Cytokinins are group of plant hormone which are isopentenyl derivatives of adenine. The different metabolites span from free N-substituted adenine base to the riboside, glucoside and nucleotide conjugates. In order to analyse all the different analytes in single run, samples were propionylated through esterification of free cytokinin hydroxyl groups by propionic anhydride. This gave a derivative with enough hydrofobicity, even of the nucleotides, to give retention on RP capillary column under acidic conditions. The use of column with high carbor load makes it possible to elute the cytokinins at a high percentage of acetonitrile (35-75%) which is favourable for the desolvation process. Furthermore a metabolites after propionylation gains a higher surface activity and thus better ionisation.
