K ověření ploidie a její stability u Quercus suber L. získaného somatickou embryogenezí byla použita průtoková cytometrie. Listové explantáty ze stromů korkovníku byly inokulovány na médium MS s 2,4-D a zeatinem. Získané embryogenní kalusy byly přeneseny na čerstvé mé-dium bez růstových regulátorů a následoval vývoj somatických embryí. Somatická embrya a listy mateřských rostlin byly použity pro určování ploidie a obsahu jaderné DNA pomocí prů-tokové cytometrie. Mezi embryi ani mezi embryi a mateřskými rostlinami nebyl zjištěn žádný signifikantní rozdíl. Také mezi regenerovanými rostlinami nebyly nalezeny žádné morfolo-gické odlišnosti.Tyto výsledky značí, že ploidie u embryo kultur a získaných rostlin je stabil-ní. Flow cytometry analyses were used to verify the ploidy stability of Quercus suber L. somatic embryogenesis process. Leaf explants of cork oak trees were inoculated on MS medium with 2,4-D and zeatin. Calluses with embryogenic structures were transferred to fresh MS medium without growth regulators and somatic embryo evolution was followed. Flow cytometry was employed to estimate DNA ploidy levels and nuclear DNA content of somatic embryos and leaves from mother plants. No significant differences (P?? 0.05) were detected among embry-os, and between the embryos and the mother plants. Also, after conversion of these embryos, no significant morphological differences were observed among the somatic embryoderived plants. These results indicate that embryo cultures and converted plantlets were stable with regard to ploidy level.
