The aim of our study was to examine in vivo and in vitro cytokines produced by Lewis rat derived R5-28 sarcoma cells. These cells produce rapidly growing tumours in approximately two weeks after subcutaneous inoculation. However, spontaneous tumour regression was noted in about 40% of animals. For an explanation of this phenomenon, we evaluated the profile of 19 cytokines during tumour growth and spontaneous regression by the use of antibody array. To detect cytokines directly originated by the sarcoma, the R5-28 cells were cultivated in vitro and then both the supernatants nad the cell lysates were analysed. Our experiments showed three cytokines to be produced by R5-28 cell in vitro. Moreover, in vivo, other three cytokines were detected both in blood sera and tumour lysates, probably produced by immune and stromal cells during tumour growth. V předložené práci bylo naším cílem charakterizovat změny v expresi cytokínů během růstu a při spontánní regresi nádorů na vlastním nádorovém modelu. Buňky linie R5-28 jsou schopné indukovat rychle rostoucí nádory v průběhu dvou týdnů po podkožní inokulaci u potkanů línie Lewis. Přibližně u 40% testovaných zvířat je možné pozorovat spontánní regresi nádorů. Úlohu imunitního systému a cytokínů jako modulátorů imunitní odpovědi na nádor jsme v tomto procesu testovali použitím „antibody array“ – techniky, umožňující simultánní detekci exprese 19 cytokínů. K rozlišení cytokínů produkovaných přímo sarkomovými buňkami R5 – 28 od cytokínů, produkovaných buňkami nádorového mikroprostředí jsme testovali expresi cytokínů v kultivačním médiu pro R5 – 28, lyzátě buněk linie R5 – 28, v celkovém nádorovém lyzátě a v séru pokusných zvířat. Naše experimenty ukázali, že buňky linie R5 – 28 produkují in vitro tři druhy cytokínů – MCP – 1, TIMP – 1 a VEGF. Typ produkovaných cytokínů koreluje s některými charakteristi...
