dcterms:description
Colostrum is a valuable food supplement as well as an interesting raw material for biotechnological processing. It contains a number of biologically active and commercially valuable components such as lactoferrin, lactoperoxidase, lysozyme, eukaryotic and prokaryotic growth factors etc. The main targets in the analysis of quality of colostrum are immunoglobulins. In industry these proteins are usually analyzed by radial immunodiffusion which is a simple, but expensive and time consuming method for the determination of immunoglobulins and other protein compounds. HPLC, as we present here, is an alternative method for the quality control of colostrum. This technique allows creating a unique elution profile of colostrum that can serve as a standard for analysis of samples obtained from different suppliers. Using HPLC the result can be obtained in just 1 hour 35 min in comparison to 24-48 h of immunodiffusion. We used two types of HPLC to study the samples of goat colostrum - reversed-phase and size exclusion chromatography. We also used radial immunodiffusion and electrophoresis in PAGE to compare our results. The results of these studies showed that HPLC is more responsive method for the quality control of colostrum comparing to immunodiffusion. We received elution profiles of cow and goat colostrum and demonstrated that reversed-phase and size exclusion chromatographies were both suitable and complement to each other for the determination of colostrum?s quality for further processing. Also these HPLC methods might be used for comprehensive characterization and separation of various valuable molecules present in cow or goat colostrum for another tests and analyses.
