The growth kinetic, overproduction of excreted recombinant protein and cell physiology of Pichia pastoris Mut+ strain were studied on mixed substrate containing two carbon sources (glucose and methanol) in chemostat. Although glucose is considered as a repressive substrate for AOX promoter, recombinant trypsinogen was expressed when glucose was used in mixture with methanol in substrate limited culture. The specific methanol uptake rate was rising with increasing dilution rate reaching maximum 0.090.01 g.gCDW-1.h-1 at 0.15 h-1; both co-substrates were utilised simultaneously and fully. At dilution rates, exceeding 0.15 h-1 the specific methanol uptake rate was decreasing and a residual methanol was detected in the culture broth. On the other hand, the specific glucose uptake rate was increasing within the interval of measured dilution rates reaching maximum (0.240.01) g.gCDW-1.h-1 at 0.2 h-1. Specificá rychlost spotřeby methanolu vzrůstala se zvyšující se zřeďovací rychlostí, maximum 0.090.01 g.gCDW-1.h-1 bylo dosaženo při D = 0.15 h-1; oba ko-substraty byly utilizovány současně a zcela. Maximální koncentrace trypsinogenu 145 mg.L-1 a maximální specifická productivita (1.17 mg.gCDW-1.h-1) byly dosaženy při D = 0.13 h-1.
