In this study, cryopreservation methods were elaborated for ex situ conservation of Bohemian tench during three experiments. Sperm was collected directly into immobilizing solution of Kurokura and equilibrated at 4°C for 2,5 h. Collected sperm was transferred to 2 ml cryotubes or straws (0.5, 1 and 5 ml) and and cryoprotectants as DMSO or mixture of DMSO with propandiol were added. Than the cryotubes /straws were directly transferred to pre-programmed PLANER Kryo 10 series III and cooled ether using a long cooling programme K4 (from +4°C to -9°C at a rate of 4°C . min-1, then from -9°C to -80°C at a rate of 11°C . min-1, held for 6 min at -80°C), or a rapid cooling programme L1 (from +4°C to -80°C at a rate of 20°C . min-1, held for 6 min at -80°C) and finally transferred into liquid N2. The spermatozoa were thwed in water bath 40°C according to the frozen volume and checked for fertilization and hatching rates. Sperma bylo odebíráno přímo do Kurokurova imobilizačního roztoku a uchováno při +4°C 2,5 hodiny. Ke spermatu byl přidán kryoprotektant, sperma bylo naplněno do 2 ml kryozkumavek nebo pejet (o objemu 0,5, 1 a 5 ml) a poté byly kryozkumavky a pejety přeneseny do předprogramovaného zmrazovacího automatu PLANER Kryo 10 serie III a mraženy delším zmrazovacím programem K4 (z teploty +4°C na -9°C rychlostí 4°C za minutu, z teploty -9°C na teplotu -80°C rychlostí 11°C za minutu, výdrž na -80°C 6 min) nebo rychlejším programem L1 ( z +4°C na -80°C rychlostí 20°C za minutu, výdrž na -80°C 6 min). Potom byly dávky přeneseny do tekutého dusíku. Sperma bylo rozmrazováno na vodní lázni teploty 40°C po dobu podle objemu dávky a použito pro test fertility na jikrách. Motilita a rychlost pohybu spermií byla zjišťována z videozáznamu mikroskopického obrazu pohybu spermií. ANOVA ve všech případech vliv zmražování.
