The durum wheat cultivars ?Cappelli? and ?Ofanto? are characterized by a clear differences in response to heat and drought stress. Wheat Affymetrix microarray was used in the two cultivars subjected to water stress at booting stage. Differential expressed genes with different stress expression profiles were found in Cappelli vs. Ofanto. According to the fold change threshold of the Affymetrix dataset and the annotated activity among these genes drought response gene markers were selected and some of them have been validated by qRT-PCR for their stabile behaving during plant development in an experiment made with two cultivars under drought stress at tillering developmental stages. These results will open the possibility to identify expression QTLs controlling the response of durum wheat to abiotic stress making possible the understanding of the genetic bases of stress dependent gene expression.
