The objectives of the study were to test diversity among winter barley genotypes exhibiting various sensitivity to FHB and to find RAPD and AFLP markers that will distinguish between susceptible and moderately resistant genotypes. A test of a set of genotypes artificially infected in field trials by fusaria was carried out. Based on the results from field and laboratory evaluation and DON content assessment, genotypes with different responses to FHB were selected. The genotypes were hybridized and DH lines were derived in F1 generation using the in vitro androgenesis method. Initial parents and derived DH lines were tested for FHB infection and DON content. Based on analyses of 80 RAPD primers in a set of parental genotypes, the primer H15 was selected that provides specific product of 650-bp size for moderately resistant. This specific product was found in 4 DH lines. A low infection level and low DON content were found in the line DH 610 from the combination of Br2611m x Duet. Cílem práce bylo ověřit diverzitu mezi genotypy ozimého ječmene s různou citlivostí k FHB a nalézt RAPD a AFLP markery, které umožní odlišení náchylných od středně rezistentních genotypů. Bylo prováděno testování genotypů v polních podmínkách, které byly uměle infikovány Fusarium culmorum. Na základě výsledků polního a laboratorního hodnocení a obsahu DONbyly vybrány genotypy s různou citlivostí k FHB. DH linie byly vytvořeny metodou androgeneze in vitro po hybridizaci mezi výchozími rodiči. Výchozí rodičovské genotypy a vytvořené DH linie byly testovány na FHB infekci a obsah DON. Na základě analýz 80 RAPD primerů v souboru rodičovských genotypů byl vybrán primer H15 poskytující specifický produkt o velikosti 650 bp pro středně rezistentní genotypy. Tento specifický produkt byl nalezen jen u 4 DH linií. Byly vyselektovány některé DH linie se zlepšenou odolnosti k FHB infekci.. Nízká úroveň napadení a nízký obsah DON byly zjištěny u linie DH 610, která vznikla z kombinace křížení Br2611m x Duet.
