Background: Macrophages are important source of cytokines and other compounds, which recruit additional cells to sites of infection or tissue injury. Importantly, their functions might be directly controlled by the composition of milieu in their immediate vicinity. Therefore, the analysis of their function under different in vitro as well as simulated in vivo condition is of particular interest. Materials and methods: Mice peritoneal macrophages were stimulated with different activators (e.g. LPS, IFN-y, TNF-a and interleukins or their combination) under different experimental conditions (in vitro and simulated in vivo conditions). Consequently, the physiological functions of macrophages were assessed using different luminometric, spectrophotometric, molecular, and immunohistochemical methods. Results: Our data demonstrate that there exists crucial difference in activation of macrophages under classical in vitro and simulated in vivo conditions.
