The principle of detection is based on enzymatic activity of alkaline phosphatase enzyme1 a component of enzyme-antibody-bacteria immunocomplex, which provides the change of enzymatic substrate (phenyl phosphate2, α ? naphthyl phosphate3) to electrochemical active product (phenol, naphthol). It?s quantified in five different buffers (carbonate, TBS2, DEA3, B-R and phosphate buffer) by voltammetric techniques (linear sweep LSV, differential pulse DPV2, square wave SWV4, normal pulse NPV5, alternate current ACV and cyclic voltammetry CV6) and amperometric method (AD1). Three electrode system is used with Ag/AgCl/3M KCl reference electrode, platinum wire auxiliary electrode and bare carbon paste electrode2,3 with 0,2mm diameter as working electrode. No stripping analysis is used, no modification of carbon paste electrode as well. Cílem práce je navrhnout nejvýhodnější kombinaci pufru a enzymatického substrátu pro amperometrickou metodu (AD) stanovení mikroorganismů na základě detekčního limitu (LOD) a meze stanovení (LOQ) pro jednoho ze zástupců patogenních bakterií v potravinách (Salmonella Enteritidis). Pro studii byly vybrány čtyři různé pufry (uhličitanový pufr, TBS, DEA, B-R) a dva enzymatické substráty (fenylfosfát, 1-naftylfosfát). Princip detekce salmonely je založen na enzymatické aktivitě enzymu alkalické fosfatázy, který je součástí imunokomplexu složeného z enzymu-protilátky-bakterie. Enzym umožňuje přeměnit enzymatický substrát na elektrochemicky aktivní produkt (fenol, 1-naftol).
