An amperometric method for salmonella detection was optimized. The method is based on the reaction of salmonella with an enzyme-linked specific antibody (linked by alcaline phosphatase), froming salmonella-antibody-alkaline phosphatase (SAAP) conjugates. After substrate hydrolysis, the product formed (phenol) are detected amperometrically using a carbon paste electrode; current signals were monited at 0.65 V vs. Ag/AgCl/sat. KCl. A medium of pH 10 was found as the best. Metoda je založena na reakci buněk salmonely se specifickou protilátkou značnou alkalickou fosfatázou. Po hydrolýze substrátu byl vzniklý fenol detekován amperometricky s využitím uhlíkové pastové elektrody. Signál proudu byl detekován při 0.65 V vs. Ag/AgCl/sat. KCl. nejvhodnějším prostedím pro detekci byl pufr s pH 10. Množství fenolu odpovídá počtu buněk Salmonella Enteritidis ve vzorku.
