It has been demonstrated in many recent studies that monitoring the T-cell population diversity is important when evaluating an individual immune status in various clinical condition, including haematopoietic stem cell transplantation, cancer, or infectious and autoimmune diseases. Analyses of T-cell receptor VB families (VB families) are most often performed to examine this diversity and the trend is to make the methodology more specific and easy to use. The aim of our study was to join this effort and to establish a method for specific and simple determination of the VB family usage. Reverse transcription PCR which allows detecting of all VB family members was combined with capillary electrophoresis in the Agilent 2100 Bioanalyzer in order to determine the relative expression values of VB families in PBMC population. Healthy donors were characterized by expression of all VB families tested, while multiple myeloma patients showed zero expression of several VB families.
