Disconnection of the peripheral axonal branch from the target tissue results in changes of GFAP expression in the satellite cells of the dorsal root ganglia (DRG) and their proliferation The loss of DRG neurons was also described after the peripheral axotomy. No results are available about the reaction of DRG neurons and their satellite cells to the central axotomy. We have investigated proliferation and apoptosis in the adult rat DRG after central and peripheral axotomy. Female rats were operated for the central axotomy and peripheral axotomy. The animals were perfused through ascending aorta with phosphate-buffered saline followed Zamboni's fixative solution after 2, 4, and 6 weeks. Cryostat sections through affected and intact DRG were incubated for immunohistochemical identification of BrdU as well as apoptosis (TUNEL). Both peripheral and central axotomy stimulated proliferation of many satellite cells while apoptosis was observed in lower amount of the cells.
