The development of a direct competitive enzyme-linked immunosorbent assay (ELISA) method based on the derivatization of semicarbazide (SEM) with o-nitrobenzaldehyde and on the immunochemical detection of the derivatited analyte is described. The small aliphatic molecule SEM was coupled to 3-carboxybenzaldehyde to produce carboxyphenyl-SEM (CPSEM), a larger hapten derivative for the generation of specific antibodies. Five rabbits produced antibodies against SEM exhibiting a 50% binding inhibition level (IC 50 value) of 0.06 to 2.28 ug /L in assay buffer. The superior indirect assay based on the antibody MVK39 showed a high dynamic range providing a linear readout in the range of 0.01 to 0.2 ug /L. The optimised ELISA based on MVK31 (IgG) was evaluated using solvent extracted SEM-spiked porcine and baby food puree samples. Práce popisuje vývoj přímé kompetititvní ELISA (enzyme-linked immunosorbent assay) metody založené na derivatizaci semicarbazidu (SEM) with o-nitrobenzaldehyde a imunonochemické detekci tohoto derivatizovaného analytu. Malá alifatická molekula SEM byla kopulována s 3-karboxybenzaldehydem za tvorby karboxyphenyl-SEM (CPSEM), který je větším haptenovým derivatem pro produkci protilátek. Pět králíků produkovalo protilátky proti SEM vykázujíce 50% vazebnou inhibici v pufru na úrovni hodnot IC 50 od 0.06 do 2.28 ug / L. Nejlepší ELISA výsledky (v nepřímém formátu) byla dosažena s protilátkou MVK39, která ukázala vysoký lineární character kalibrace v rozsahu koncentrace 0.01 to 0.2 ug / L. Optimalizovaná ELISA pracující s MVK31 (IgG) protilátkou byla evaluována s použitím fortikovaných extraktů vzorků z hovězího masa a dětské výživy.
