We will be concetrated on pig oocytes maturation in vitro, their artificial activation and their subsequent invasive enucleation by mikromanipulation.Development of an effective activation and invasive enucleation protocols is of great importance for studying oocytes competence and embryo cloning. We would like to also try the noninvasive chemical enucleation, because this method of enucleation hasn´t been developed in pigs yet. This novel enucleation technique could greatly facilitate the research in pig cloning experiments. Althrough the birth of the first lamb produced by nuclear transfer of differentiated cell into the enucleated metaphase II oocyte has been reported, the success of current cloning procedures is still extremely low for unknown reasons. We do not know if the differentiation of karyoplasts and our inability to reprogramme them is the main reason why the development of clones is so poor. Another factor, which may well be involved is the synchronization of cell cycles between karyoplasts and cytoplasts. The purpose of our cooperation project is the detailed study of nucleo-cytoplasmic interactions in reconstructed embryo and the effect of different combinations upon nuclear reprogramming and futher development.
