Hlavním cílem projektu je pripravit v systému in planta standartní a modifikované varianty klonovaných bifunkčních nukleáz, vybrané alternativy analyzovat z hlediska katalytických, strukturních, antiproliferativních, biologických aktivit a mechanismu účinku u zvolených živočišných modelů. Previously we discovered anticancerogenic effects of plant nucleases I. Antiproliferative effects were reached at protein concentrations approximately ten-times lower in comparison to studied animal RNases and side-effects were much lower than caused by animal RNases. This makes plant nucleases perspective anticancerogenic agents that could be, similarly to onconase, selected for clinical trials in the future. In the project proposed we aim to study modifications and biological properties of three cloned nucleases, TBN1 from tomato, HBN1 from hop and ABN1 from A. brassica.. Site-specific mutagenesis, cDNA shuffling and posttranslational modifications in planta are proposed to study structural-functional relationships responsible for nuclease activity, stability and for mechanism(s) of antiproliferative effects that will be analyzed in detail especially in human melanoma, and sassayed using other selected cancer models. Levels of immunosuppression, aspermatogenesis and effects on blood formation will be estimated to characterize potential side-effects of nucleases produced by in planta biotechnology.
