Sonic hedgehog (Shh) and Wnt signalling are both required for the maintenance of stem cell niches in the postnatal subventricular zone and for the proliferation of neural stem/progenitor cells in the mature hippocampus. The aim of this project is to elicidate the effect of two morphogenes, Shh and Wnt-7a, on neuro- and gliogenesis using neural stem/progenitor cells that express either Shh or Wnt-7a. Their role in neural stem/progenitor proliferation and differentiation will be studied in vitro and after cell transplantation into the site of ischemic lesions in the rat brain. In addition to endogenous neural stem cells, also astrocytes and NG2-glia play an important role in neuro- and gliogenesis during ischemia/reperfusion. Their proliferation, migration as well as the proliferation and differentiation of GFP-labeled endogenous neural stem cells, will be investigated in response to the stereotactic delivery of morphogenes and growth factors into the lateral ventricles, using immunohistochemical and electrophysiological techniques. Our data will provide information about the role of morphogenes in brain regeneration after ischemia/ reperfusion in connection with the proliferation and differentiation of neural stem/progenitor cells as well as the proliferation and migration of astrocytes and NG2-glia. This study should also contribute to the development of new therapeutic approaches to the treatment of cerebral ischemia. Cílem projektu je studovat regeneraci ischemicky poškozené nervové tkáně, se zaměřením na vliv morfogenů (Sonic Hedgehog a Wnt-7a) a růstových faktorů na proliferaci, migraci a differenciaci endogenních neurálních kmenových buněk, a úlohu NG2-glií a reaktivních astrocytů v neurogenezi a gliogenezi.
