Vytvoření a analýza transgenních SHR potkanů s lidským C-reaktivním proteinem s definovanou tkáňovou a kvantifikovatelnou expresí umožní odhalit úlohu CRP v patogenezi metabolického syndromu a preklinické testování nových terapeutických inhibitorů CRP. We propose to create advanced transgenic rat models in which tissue specific expression of human C-reactive protein (CRP) can be reversibly and quantitatively controlled in a manner that will: a) enable investigators to directly study mechanisms whereby variations in tissue and circulating levels of human CRP influence pathogenesis of the metabolic syndrome and related inflammatory cardiovascular disorders and b) facilitate the preclinical testing of new CRP inhibitors for use in the prevention and treatment of diabetes and cardiovascular disease. The derivation of such as transgenic lines will be provided using Te-On regulatory system. 1) Specific DNA constructs will be designed and synthesized and the transgenic SHR lines will be derived. SHR transgenic line with the Tet response element (TRE) – human CRP construct will be crossed with SHR transgenic line expressing the reverse tetracycline activator (rTA) under the control of tissue specific promoter.
